Porous β-tricalcium Phosphate Schaffolds with Sustained Therapeutic Mg Ions Release

Muskoleskeletal diseases or disorders such as osteoporosis, osteonecrosis and degenerative osteoarthritis affect hundreds of millions of people across the world. These bone diseases often lead to bone fragility and complex fracture. These diseases often demand a drug treatment at the specific surgery, injury or defect site. Different therapeutic agents such as antibiotics, anticancer, anti-inflammatory or growth factors, bioactive proteins, enzymes are used in bone tissue engineering to induce osteoinductivity in the implanted biomaterials to accelerate the healing and regeneration process of bone tissues. Calcium phosphate (CaP) bioceramic scaffolds are excellent choice for bone tissue engineering for bone replacement, augmentation and regeneration. In an ideal case, CaP scaffolds would be macro and microporous and can eventually degrade while the newly formed bone tissue takes over the space. β-Tricalcium phosphate (β-TCP) bioceramics are bioactive and highly bioresorbable in bone tissues. Mg is one of the most important metallic ions, which affects all stages of skeletal metabolism and play a pivotal role of the initial stage of osteogenesis in cartilage and bone tissue. Mg deficiency has been suggested to be a potential risk factor for osteoporosis. Mg incorporation into β-TCP crystal structure reduces crystallinity and therefore bioresorption activity in the body. Therefore, Mg containing porous β-TCP scaffold is excellent bioceramic matrice for sustainable and therapeutic Mg ions release. The main purpose of this work was evaluated the self-dissolution behavior in vitro of hierarchically 3D microporous-macroporous β-TCP scaffolds containing various amounts of Mg.
